Description
Liraglutide — Acylated GLP-1 Receptor Agonist Peptide
Liraglutide is a synthetic acylated analogue of GLP-1, engineered with a fatty-acid side chain to extend plasma protein-binding, acting as an agonist at the GLP-1 receptor. It is supplied for in vitro laboratory research use only.
Research context
Published literature has examined liraglutide’s binding affinity and cAMP-accumulation activity at the GLP-1 receptor in transfected cell-line systems, including pancreatic beta-cell line model systems used to characterise receptor-mediated signal transduction. Structure-activity studies have examined the C16 fatty-acid acylation that confers extended plasma albumin-binding and altered proteolytic-degradation kinetics relative to native GLP-1. Receptor-binding and functional cAMP-signalling assays remain the primary in vitro methodologies applied to this compound in the literature. Receptor-internalisation and desensitisation assays in transfected cell lines have additionally been used to characterise prolonged receptor-occupancy behaviour associated with the acylated structure, using fluorescence-based receptor-trafficking assay formats in transfected reporter cell lines.
