Description
AICAR — AMPK-Activating Nucleotide Analogue
AICAR (5-aminoimidazole-4-carboxamide ribonucleotide) is a cell-permeable nucleotide analogue of adenosine monophosphate, engineered to activate AMP-activated protein kinase (AMPK). It is supplied for in vitro laboratory research use only.
Research context
Published biochemical literature has examined AICAR’s intracellular conversion to ZMP and its downstream allosteric activation of AMPK, characterised using kinase-activity and phosphorylation-state assays in cultured cell-line systems. In vitro studies in isolated myotube and hepatocyte cell cultures have examined AMPK-pathway effects on glucose-transporter trafficking and fatty-acid oxidation enzyme activity at the cellular level. Kinase-activity assays and cell-based AMPK-pathway reporter systems remain the primary in vitro methodologies applied to this compound. Metabolic-flux assays using radiolabelled glucose and fatty-acid substrates have additionally been used to quantify pathway activity in these cell-culture systems following AMPK activation, with parallel immunoblot analysis of downstream phosphorylation targets across multiple independent cell-line passages and several incubation time points.
